《Harmful algae》2015
Cyanobacteria blooms may present a public health concern in sources of drinking water and recreational water due to the production of toxins by some species, microcystins being the most commonly found. It is possible to detect microcystins using instrumental analyses and field test kits. While instrumental analysis methods are accurate, they are also costly, and in regions with a high incidence of blooms the time to report is lengthy (days). On the other hand, the use of commercially available test kits may provide quicker results at a lower cost. The purpose of this work was to evaluate three commercially available kits: the Immunochromatographic Strip Test for the Detection of Microcystins and Nodularins in Source Drinking Water at 1 μg/L (Abraxis strip test), the Abraxis Microcystin Tube Kit and the Envirologix QualiTube Kit. The evaluation of each kit focussed on the interpretation of the results by the end-user and the validity of a test kit was based on four indices: sensitivity, specificity, positive predictive rate (PPR) and negative predictive rate (NPR) (false positive/negative) based on the manufacturer's specifications. The results indicate that there are challenges in the visual interpretation of the results at levels close to the threshold value for each kit. The scope of each kit must be understood: free vs. total, qualitative vs. semiquantitative. For instance, the Envirologix Qualitube Kit does not provide a lysing agent, therefore it will underestimate the levels of total microcystin if a lysing step is not included. In the case of the Abraxis strip test, the kit provides information on the absence/presence of microcystin at a threshold value of 1 μg/L, but false positives were encountered. 相似文献

A series of novel HIV-1 protease inhibitors has been designed and synthesized, which contained morpholine derivatives as the P2 ligands and hydrophobic cyclopropyl as the P1′ ligand at the meantime in this study, with the aim of improving the interactions between the active sites of HIV-1 protease and the inhibitors. Twenty-eight compounds were synthesized and assessed, among which inhibitors m18 and m1 exhibited excellent inhibitory effect on the activity of HIV-1 protease with IC50 value of 47 nM and 53 nM, respectively. The molecular modeling of m1 revealed possible hydrogen bondings or van der Waals between the inhibitor and the protease, worthy of in-depth study. 相似文献

The nature and persistence of seed quality effects produced by a range of seed production treatments was investigated in the autumn-sown onion crop. Genotypically equivalent seed lots raised in different glasshouses within the same year were found to germinate at significantly different rates. The seed lot which emerged earlier produced larger seedlings and this difference in size persisted throughout the growing period. In a comparison between the effects of hand crossing and self-pollination techniques the hand crossing method produced a much lower total seed yield but heavier seeds and larger plants. The induced effects persisted throughout the year but were not significant in the mature bulb characters. The implications of these results upon the control of seed production for plant breeding and genetical experiments is discussed. 相似文献

Zusammenfassung Die serotoninhaltigen, nach Formaldehydbedampfung im UV-Licht gelbfluoreszierenden EC-Zellen im Magendarmepithel wurden bei normalen und bei tryptophanfrei ernährten Ratten untersucht. Bei Kontrolltieren ist die Zahl der EC-Zellen, bezogen auf die Schleimhautfläche, im Duodenum größer als im Pylorus. Im Duodenum sind die EC-Zellen etwa gleichmäßig auf Oberflächen- und Drüsenepithel verteilt, im Pylorus ganz überwiegend im Drüsenepithel lokalisiert. Die Brunnerschen Drüsen besitzen keine EC-Zellen. Tryptophanfreie Diät führt zu starker Abnahme des Serotoningehalts der EC-Zellen, nach Umsetzung auf Normalkost steigt er wieder an. — Die Ergebnisse sprechen weiterhin dafür, daß die EC-Zelle sowohl Speicher- als auch Syntheseort von Serotonin ist. Dabei kommt Serotonin in der EC-Zelle wahrscheinlich in zwei Fraktionen vor, nämlich zytoplasmatisch gelöst und granulagebunden. Die histochemischen Nachweismethoden für Serotonin, Möglichkeiten des Eingriffs in die verschiedenen Syntheseschritte des Serotonins und deren Spezifität in bezug auf die EC-Zelle als physiologischem Ort der Serotoninsynthese werden diskutiert.

Summary The serotonin-containing EC-cells of the gastro-intestinal mucosa, which show yellow fluorescence after treatment with gaseous formaldehyde, were investigated in the rat after normal and tryptophan-free nourishment. In control animals the number of EC-cells, related to epithelium area is higher in the duodenum than in the pyloric region. In the duodenum the EC-cells show about the same distribution in surface epithelium and gland epithelium. In the pyloric region EC-cells are localized predominantly in the gland epithelium. No EC-cells were found in the Brunner glands. After tryptophan-free diet the serotonin-level of the EC-cells strongly decreases; serotonin-level increases after return to normal nourishment. The results also suggest, that serotonin is synthesized as well as stored in the EC-cell and that it occurs in two fractions: bound to granules and dissolved in the cytoplasm. Discussion deals with the histochemical methods for determination of serotonin, the possibilities of influencing the pathways of serotonin metabolism and the specificity of these possibilities concerning the EC-cell as the physiological site of serotonin synthesis.

Summary A quantitative evaluation has been made of the sizes of neuronal granular vesicles (GV) in different (sub-)zones of the rat
median eminence. On the basis of the frequency distribution of the mean sizes of the GV in the individual nerve profiles,
at least 4 tentative categories of nerve profiles may be distinguished. However, available data do not permit characterization
of nerve fibre categories in terms of neurohormone and/or neurotransmitter storage.
From dorsal to ventral the palisade zone shows a decrease in mean size of GV; furthermore, the relative number of nerve profiles
characterized by small GV (mean size smaller than 112 nm) increases. In contrast, the GV in the zona granulosa, that part
of the external zone of the median eminence ventral to the palisade zone, are of relatively large size.
Qualitative observations indicate that the zona granulosa may be regarded as an area for both storage and release of biologically
active substances.

A series of novel N-alkylated C-6-isobutyl- or -propyl pyrimidine derivatives were synthesized and their antiproliferative effect was evaluated on a panel of tumor cell lines including leukemia cell line K562 and normal diploid human fibroblasts. N-methoxymethylated 5-methylpyrimidin-2,4-dione with di(benzyloxy)isobutyl at C-6 (14b) showed the strongest effect on the cell growth at micromolar concentrations. Mechanisms of action for the lipophilic compound 14b predicted in silico, pointed to its anticancer and antimetastatic potential exerted through inhibition of DNA or RNA polymerases and adhesion molecules. Global biodiversity is facing an extinction crisis. Australia has one of the highest terrestrial species extinction rates in the world. Scientists, policy advisors and governments have recommended that the issue be addressed at a landscape-scale, while noting that there are significant knowledge gaps that are hampering implementation of such an approach. From 2011–2015, the Australian Government funded a transdisciplinary research program, the Landscapes and Policy Hub, to meet this need. Transdisciplinary research is widely acknowledged as essential to address the complexity of contemporary environmental problems. Given that such research programs are in their infancy, it is important to evaluate their efficacy and provide an empirical basis for improving their design. This paper presents an evaluation of the strategies fostering transdisciplinarity adopted by the Landscapes and Policy Hub. A heavy emphasis on communication, with skilled knowledge brokering, regular face-to-face meetings using participatory activities and shared field engagements enhanced transdisciplinary interaction between researchers and research users. However, establishing a fully integrated interdisciplinary research program remained a challenge. Efforts to enable shared conceptual frameworks to emerge through adaptive application of theory in practice could have been balanced with increased effort at the outset for researchers and research users to collaboratively formulate shared research questions, leading to the establishment of teams that could address these questions through cross-mobilisation of interdisciplinary expertise. 相似文献

Environmental management, being an important component in strategies for achieving sustainable development of processes and products, has emerged as a proactive approach in majority of the manufacturing organizations. From the strategic perspective environmentally conscious manufacturing (ECM) programs lead to better environmental management practice. The objective of the current paper is to present an integrated and holistic framework to evaluate ECM programs. This framework combines three multi-criteria decision analysis (MCDA) methods to consider eight major environmentally conscious manufacturing indicators (ECMI) in order to identify the efficiency of each ECM program. First the interdependence relationship among the ECMIs is established using decision-making trail and evaluation laboratory (DEMATEL). Then a range of weightage (i.e. upper and lower bounds) is created for each ECMI using analytic network process (ANP) to include managerial preferences. Finally, this range of weightage for each indicator is applied to perform restricted multiplier data envelopment analysis (RMDEA). Results show that the technical efficiency of the inefficient ECM programs for integrated RMDEA, on average, is calculated as 53.2% whereas traditional input oriented DEA provides the same score as 72.3%. This clearly indicates that integrated RMDEA is better than the input oriented DEA because same level of output could be produced with lesser resources if the ECM programs perform on the frontier. Hence, the advantage of this methodology is that the managerial preferences are successfully implemented through this newly developed hybrid methodology that will help to reduce less resource consumption and lead to better environmental policy. 相似文献

资源型城市作为我国重要战略保障基地,面临较为突出的不可持续问题,定量化开展可持续发展水平监测评估是推动其绿色转型发展、激活再生动力的重要途径。基于联合国可持续发展目标体系(SDGs),结合行星边界理论,从资源型城市可持续发展特征出发,建立了由2个系统、4个支柱、12个领域和48项指标组成的资源型城市可持续发展评价指标体系。借鉴全球SDGs监测评估实践经验,考虑指标属性,综合确定各指标的阈值,并采用改进的离差标准化、障碍度分析及耦合协调度分析等方法建立综合评价技术体系。以拥有“有色金属之乡”称号的郴州市国家可持续发展议程创新示范区为评价对象,对其2005—2019年连续15年的可持续发展水平进行实证分析。结果表明:(1)郴州市可持续发展水平整体上呈现稳步上升趋势,得分由0.398上升到0.610。(2)4个支柱中,自然资源支柱的得分进步幅度最大,高达71.3%,说明郴州市已经逐步提高对资源利用效率的重视,同时意识到挖掘替代资源的重要性,资源保障与利用能力不断提升;而发展动力支柱得分始终处于较弱或中等水平,已成为制约郴州市可持续发展和绿色转型的主要因素,其中科技创新驱动是该支柱发展关键障碍... 相似文献
